The processes of clotting and lysis then were monitored over time by nrrbidity. The results showed that, under the conditions employed, the time required for the clot to lyse progressively lengthened from 30 min in the absence of TAFIa to about 90 min a1 10rM TAFIa (19). The phenomenon is saturable in TAFIa, and the half maximal effect was obtained at a TAFIa concentration of 1.0 nM. Since the TAFI concentration in plasma is about 75 nM, sufficient TAFIa clearly can be generated from the precursor to substantially perturb fibrinolysis. Other studies (37) showed that when fibrin clots are lysed in the presence of TAFIa, stoichiometric quantities of both arginine and lysine are released, in accordance with the conclusion that TAFIa catalyzes removal of the lysine and arginine residues from the carboxy termini of chains that appear within fibrin following plasmin catalyznd bond cleavage.
