MATERIALS AND METHODS Animal Exposures and Tissue Preparation: Female C57BL6
wild-type and NOS knockout mice (n= 2/group) were exposed to either normoxic (21% O 2)
or hypoxic (10% O 2) conditions for 3 weeks. After exposure, the mice were anesthetized
and the lungs perfused through the pulmonary arteries with 2 to 4 mL of heparinized saline.
The lungs were excised, flash frozen in liquid nitrogen, and stored at− 70 C.
