Reports the effects of clozapine (CLZ) on 2 8-yr-old boys and 1 12-yr-old girl, all with autistic
disorder. CLZ was administered at progressively increasing doses and then modified
according to clinical response. After 3 mo, CLZ treatment markedly decreased scores on the
Children's Psychiatric Rating Scale (CPRS) for all 3 Ss (from 99, 99, and 81 to 60, 80, and

After 3 mo, CLZ treatment markedly decreased scores on the Children's Psychiatric Rating Scale (CPRS) for all 3 Ss (from 99, 99, and 81 to 60, 80, and 53, respectively). The boys continued to improve: hyperactivity and blunted affect were markedly reduced, and a substantial improvement in language and communicative skills was evident. The girl, however, once again began to display self-injurious behavior, hyperactivity, and stereotypies.
