Janus (Jak) tyrosine kinases contain a tyrosine kinase (JH1) domain adjacent to a
catalytically inactive pseudokinase domain (JH2). The JH2 domain has been implicated in
regulation of Jak activity, but its function remains poorly understood. Here, we found that the
JH2 domain negatively regulates the activity of Jak2 and Jak3. Deletion of JH2 resulted in
increased tyrosine phosphorylation of the Jak2-and Jak3-JH2 deletion mutants as well as of
