FK-506 (FK) is a novel immunosuppressant isolated from Streptomyces tsukubaensis (Fig
1). It is nearly 500 times more potent than cyclosporine (CyA) in inhibiting lymphocyte
proliferation in mixed lymphocyte cultures. 1 In vivo, it significantly prolongs the graft survival
in rats receiving heterotopic heart transplants2 and dogs receiving renal3 or liver
transplants. 4
