The establishment and maintenance of immunological tolerance entails both central and
peripheral mechanisms. The latter have been highlighted in the past several years, mostly
because of great interest in the activities of regulatory T cells. However, an important role for
central tolerance mechanisms has been reemphasized by recent results on human
autoimmune diseases, including APECED and type 1 diabetes.
