The chemical structure of cord factor of Mycobacterium tuberculosis has been established as
trehalose-6, 6'-dimycolate (I), C 186 H 366 O 17±10CH 2. Alkaline hydrolysis of cord factor
yields two molecules of mycolic acid and one molecule of the disaccharide trehalose.
Methylation studies show that the two mycolic acid molecules are attached through an ester
linkage to the primary hydroxyl groups of trehalose. Identical results were obtained with cord
factor from the human strain Brévannes, and the BCG strain.
