Summary [3H] palmitate attached to mutationally activated a.(aA3201C) turns over rapidly, compared with palmitate linked to normal a.(tv,= 2 min versus 90 min); although a,-R2OlC (unlike normal a,) is predominantly found in the cytosol, PH] palmitate is linked only to the membrane-bound pool of a,, normal or mutant. Similarly, activation of wild-type Q by isoproterenol, a 9-adrenoceptor agonist that also induces membrane-to-cytosol translocation of u., dramatically accelerates depalmitoylation of G. Thus, activation-induced removal of palmitate provides an explanation for actlvationinduced shifts of a, to the cytosol. Regulated palmitoy lation cycles provide a potential general mechanism for controlling reversible changes in the cellular location and activity of a protein.
