Effect of activation of NF-κB on COX2 expression. A constitutive active IKKα was transduced into cultured rabbit RMICs by an adenoviral vector, COX2 immunoreactive protein and COX2 mRNA expressions were examined. (a) Immunoblot of COX2 before and 12, 24, 36, and 48 hours after transduction of IKKα. Successful IKKα transducing was examined by immunoblot using anti-IKKα Ab (lower panel). (b) Dominant-negative IκB was transduced into RMICs together with constitutive active IKKα, and their effect on COX2 expression was determined 48 hours later with immunoblot. (c) Nuclease protection assay showing COX2 mRNA expression in cultured RMICs transduced with the IKKα or IKKα together with IκBmut.
