Cells of erythroid lineage express EPO mRNA. (a) CB CD34+ cells were cultured in the presence of SCF and sIL-6R/IL-6 for 14 days and the cells were FACS sorted into erythroids (R1), myeloids (R2), and other cells (R3) using anti–hGPA-FITC and anti–hCD13-PE mAb’s. (b) cDNAs were synthesized from sorted cells after RNA extraction, and EPO mRNA detection was carried out using RT-PCR (40 cycles). Expression of EPOR, GPA, and β-actin mRNAs was also examined by RT-PCR (33 cycles). Size of the markers is indicated on the left.
