Effect of AQP5 deletion on active alveolar fluid clearance. Measurements were done at 37°C using an in situ perfused lung preparation in which the airspace was instilled with an isosmolar solution containing 131I-albumin as a volume marker. Alveolar fluid clearance is expressed as percentage fluid absorption (mean ± SD, n = 6–10 mice) at 15 minutes. (a) Measurements of lungs from wild-type mice. Where indicated, the instillate contained isoproterenol (0.1 mM), terbutaline (10 mM), amiloride (1 mM), or ouabain (0.1 mM), or measurements were performed 72 hours after treatment of mice with KGF (see Methods). (b) Measurements on lungs from litter-matched wild-type mice and AQP5 knockout mice. Data from individual mice (filled circles) and averaged (mean ± SD) values are shown. The instillate contained 1 mM isoproterenol in all experiments. Where indicated, mice were pretreated with KGF.
