IL-1 and TNF induce TNF gene expression via the TRE site in the TNF promoter. Raw 264.7 cells were transiently transfected with either a wild-type TNF/CAT construct containing the 2.2-kb 5′ promoter region plus 1 kb of the 3′UTR or the corresponding plasmid with a site-directed mutation of the TRE site. Cells were then stimulated with either IL-1 + TNF or LPS for 12 hours. CAT activity was normalized to β-galactosidase activity to correct for variability in transfection efficiency and expressed as normalized CAT activity. Mean ± SEM of 3 experiments. *P < 0.05 vs. unstimulated cells. **P < 0.01 vs. cells transfected with the wild-type TNF/CAT reporter gene construct.
