E2 decreases TNF gene expression in a dose-dependent manner. Raw 264.7 cells were pretreated with E2 (10–7 to 10–12 M) for 24 hours before transfection and then transiently transfected with either a TNF/CAT construct containing the 2.2-kb 5′ promoter region plus 1 kb of the 3′ UTR (filled bars) or a TNF/CAT construct containing the 2.2-kb 5′ promoter region lacking the 3′ UTR region (open bars). The cells were then treated again with E2 (10–7–10–12 M) and 5 ng of TNF and IL-1 for 12 hours. Mean ± SEM of 4 experiments. *P < 0.05 vs. untreated cells.
