S-nitrosylation of pVHL mimics hypoxia.

pVHL is essential for the ubiquitination and rapid degradation of HIF-α. Posttranslational hydroxylation of proline residues in the oxygen-dependent degradation (ODD) domain of HIF-α is required for the interaction between HIF-α and pVHL. This requires molecular oxygen. pVHL is a target for S-nitrosylation. HIF-α and pVHL fail to interact, and the degradation of HIF-α is impaired. Therefore, even in the presence of oxygen, the cells perceive a hypoxic stimulus. Ub, ubiquitin.
