Either blockade of β1 integrin signaling with neutralizing antibody or knockdown of its expression abolishes the tPA-mediated induction of myofibroblast activation.

(AC) NRK-49F cells were pretreated with mouse IgG or β1 integrin antibody (12.5 μg/ml) for 1 hour, followed by treatment with tPA in the absence or presence of TGF-β1 for 24 hours. Blockade of β1 integrin signaling by neutralizing antibody eradicated the tPA-mediated induction of α-SMA (A) and type I collagen (B and C) in NRK-49F cells. Quantitative determination of type I collagen mRNA levels in different groups is presented in C. *P < 0.05. (D) Knockdown of β1 integrin also abolished the tPA-mediated induction of myofibroblast activation. NRK-49F cells were transfected with β1 integrin–specific siRNA or control siRNA, followed by incubating with tPA in the absence or presence of TGF-β1. Knockdown of β1 integrin expression was confirmed by western blot analysis.
