Schematic overview of enhanced immunoproteasome-mediated NF-κB activation and consequent exacerbation of inflammation in CD patients. Unknown initial inflammatory events induce expression of c-Rel in the intestinal mucosa and a strong Th1 cell response via IL-12. High amounts of IFN-γ trigger generation of immunoproteasomes, which enhances NF-κB activation, thus perpetuating the inflammatory process.
