Therapeutic effects of TACI:Fc in aggregate synovitis. Human synovium–SCID mouse chimeras implanted with synovial tissues with aggregates were treated with TACI:Fc or control Fc and then analyzed as described in Figure 6. Results from experiments with tissues from 5 patients are shown. Blocking APRIL/BLyS left production of IgG1/2 transcripts totally unaffected. Following treatment with TACI:Fc, IFN-γ production markedly increased. DC function assessed through CCL19 remained unchanged. Transcription of TNF-α was significantly diminished. Results are given as box plots, as described in Figure 1.
