Effect of resistin on Akt phosphorylation in skeletal muscle, liver, and adipose tissue. Basal and insulin clamp–stimulated skeletal muscle, adipose tissue, and liver samples were obtained and homogenized as described in Methods. After fractionation of the resulting homogenates (50 μg) by 10% SDS-PAGE, proteins were transferred onto PVDF membranes. Membranes were probed with phosphorylated Akt (pAkt) (Ser473 and Thr308) and Akt antibodies. The bar graphs show data quantification by NIH Image for results in skeletal muscle, adipose tissue, and liver. Values are means ± SEM of three independent experiments using six animals for each group. Data are expressed relative to control Adv-lacZ values, with a value of 100% assigned to the insulin-stimulated control mean. *P < 0.01.
