Induction of IL-12 production by DCs is dramatically increased by Th cells stimulated with higher-affinity peptide and is blocked by anti-CD40L. Triplicate cultures of 5 × 104 T1-specific CD4+ Th cell clone were stimulated with 5 × 105 DCs, and 50 hours later culture supernatants were assayed for IL-12 by ELISA. These results were highly reproducible in three separate experiments.
