The pattern of insulin-induced membrane ruffling parallels the immunostaining of GLUT4myc on the surface of nonpermeabilized myotubes. Serum-deprived L6 GLUT4myc myotubes were stimulated with 100 nM insulin for 10 minutes at 37°C, then stained with anti-myc antibody followed by FITC-conjugated goat anti-mouse antibody to detect cell-surface GLUT4myc, as described in Methods.
